Johnny Hooker, a thief in Joliet, Illinois, con $11,000 in cash from an unwary victim in 1936, with the help of his partners Luther Coleman and Joe Erie. Buoyed by the windfall, Luther declares his retirement and advises Hooker to seek out an old friend, Henry Gondorff, in Chicago to learn “the big con”. Unfortunately for them, their prey was a numbers racket courier for vicious crime kingpin Doyle Lonnegan. Hooker is confronted by corrupt Joliet police lieutenant William Snyder, who reveals Lonnegan’s participation and demands a portion of Hooker’s cut. Hooker pays Snyder in counterfeit bills after blowing his share on a single roulette roll. Hooker escapes to Chicago for his life after Lonnegan’s men murder both the courier and Luther. Hooker tracks down Gondorff, a once-famous con man who is now hiding from the FBI, and begs for his assistance in taking on Lonnegan. Gondorff is initially hesitant, but eventually relents and gathers a squad of experienced con artists to dupe Lonnegan. They plan to resurrect the elaborate, defunct “wire” scam, enlisting a larger team of con artists to set up a phony off-track betting parlor. Gondorff, posing as a snobbish Chicago bookie named Shaw, buys into Lonnegan’s secret, high-stakes poker game aboard the opulent 20th Century Limited. He irritates Lonnegan with his obnoxious conduct before defrauding him of $15,000. Hooker is sent to collect the winnings as Shaw’s disgruntled employee Kelly, but instead convinces Lonnegan that he wants to take over Shaw’s business. Kelly reveals that he has a partner in the Chicago Western Union office named Les Harmon (actually con man Kid Twist) who will assist them in deposing Shaw by winning bets he makes on horse races through past-posting.

Snyder has traced Hooker to Chicago, but his pursuit is thwarted when he is called by undercover FBI agents headed by Agent Polk, who orders him to assist in the capture of Gondorff. Simultaneously, Lonnegan has become dissatisfied with his men’s inability to locate and kill Hooker for the Joliet con. Unaware that Kelly is Hooker, he insists that the job be given to Salino, his best assassin. Hooker is followed and observed by a mysterious person wearing black leather gloves. Polk then shoots Gondorff and instructs Snyder to remove the seemingly reputable Lonnegan from the crime scene. Hooker and Gondorff rise amid cheers and laughter now that Lonnegan and Snyder have been securely removed. Polk is actually Hickey, conducting a con within a con to divert Snyder and ensure that Lonnegan abandons his money. Hooker rejects his share of the money and walks away with Gondorff as the con men empty the room.
